Mbuemo converted and extended his goalscoring run to four consecutive top-flight games. Rico Henry's surge down the left-hand side led to Wissa's goal. Royal scored the pick of the goals with a guided finish in the fifth minute of 11 added on.
Tottenham will have to make do without key forward Toney until mid-January after he was banned for eight months for breaking Football Association betting rules.
Tottenham kicked off their post-Harry Kane era with an eventful draw against Brentford in the Premier League's opening weekend.
After Harry Kane's departure to Bayern Munich was confirmed on Saturday, new Spurs manager Ange Postecoglou secured a point in Sunday's match through goals from Christian Romero and Emerson Royal.
Brentford, missing their key player Ivan Toney, managed to take the lead as Yoane Wissa scored following Bryan Mbeumo's successful penalty kick, which had canceled out Romero's opener.
Spurs have now experienced three winless outings at the Gtech Community Stadium. The match's start was delayed by six minutes due to a water supply issue.
Argentina's defender, Romero, opened the scoring with a precise header from James Maddison's powerful free-kick, marking his last contribution before being substituted as a precaution following a knock to the head during the celebration.
Brentford responded by earning a penalty after Son Heung-min, Spurs' new captain, brought down Mathias Jensen, allowing Mbuemo to convert from the spot and extend his streak of four consecutive top-flight games with goals.
Wissa then capitalized on Rico Henry's rapid run down the left flank, slotting home the cutback past Guglielmo Vicario with the help of a deflection off Micky van de Ven's boot.
Emerson Royal added a remarkable goal with a guided finish during the 11 minutes of added time at the end of a thrilling first half.
Brentford knows they'll have to cope without key striker Toney until mid-January due to an eight-month ban for breaching Football Association betting regulations.
However, the team appears unfazed by his absence, as they had previously won four of five games without him last season. With Mbeumo and Wissa as capable forwards, Brentford continues to demonstrate their attacking prowess.
Despite Kane's departure, Tottenham's fans are rightfully concerned about the void he left behind in terms of goalscoring. Kane, with 280 goals in 435 appearances, was their all-time top scorer.
Although Brazil's Richarlison appears to be Postecoglou's initial choice to fill that void, he struggled to ease the fans' worries. In the first half dominated by Spurs with 66% possession, Richarlison only touched the ball 16 times and remained relatively isolated.
Even Maddison, who was a significant summer signing from Leicester, struggled to set up Richarlison effectively. The combination attempted to create opportunities, but Richarlison's shot was straight at Burnley's goalkeeper Mark Flekken.
Time will reveal whether Richarlison can become Tottenham's long-term solution to Kane's departure.
