Lucas Paqueta converted a penalty to make up for his role in the opponents' 26th-minute goal, and Michail Antonio scored twice.
Declan Rice, the team's captain, added a stunning third goal by scoring with a low shot at the conclusion of a ferocious run that began in his own half.
Gent, who had taken the aggregate lead thanks to a goal from Hugo Cuypers in the first half, were ultimately outmatched.
'That's why we value Rice so highly' - Moyes
For only the second time in West Ham's European history, Jarrod Bowen had a goal disallowed for handball.
They currently have a sixth semi-final to look forward to and yet have a chance to follow in the footsteps of the European Cup Winners' Cup-winning team from 1965. They can also end a significant trophy drought that dates back to 1980 and their FA Cup victory over Arsenal.
The manager, David Moyes, will be fully aware that Premier League survival would have to go hand in hand with European victory, but it also means that what has at times been a tortuous season may conclude in triumph.
